超出现有显微镜的成像极限
大多数3D显微成像技术是通过对样品的位置深度进行扫描来实现;由于生物样品动力学研究和对光敏感的苛刻限制,Insight采用非扫描技术,实现单次3D成像采集,使样品尽量避免光毒性或光漂白,从而确保在高时间分辨率的条件下采集3D动态事件。
工作原理
不同于传统扫描系统扫描样品时所需要移动的时间限制,Insight采用非样品扫描方式,该技术基于高分辨率光场成像技术和PhaseView采集和软件引擎。
Phaseview光场成像存储空间和角度信息,而且不会向典型的小透镜阵列损失个别性能。
这种新方法集合了可变焦距的镜头和微透镜整列。如下图所示:
4D的数据将通过Phaseview的软件进行处理和分析。
